The third reason is the changes of personality. In some families, parents' divorce and family members' death could leave bad memories to their children. After that, people will have more negative emotions and they will think that lives are too hard to handle. So they may not want to talk to their friends about those bad emotions. As a friend, if people do not want to talk to me, I will think that he doesn’t want to be my friend any more. Then misunderstandings come out, and friendship not longer lasts. I think after we've grow up, people’s habits may change a lot. So people`who like different things may enjoy different lives. There is a gap existing if friends enjoy different things. And friendship may not last.
Those three reasons are the most important reasons for me that could bring friendship to an end. But I know that there are still some pacific reasons which could lead to situation. my life experience tells me that a good friendship might be difficult in some less important aspescts but it can still last for a long time. Treasure the friends with whom people have and enjoy livese, there will be more benefits from lives.

i am writing this log to record down what and how i had done in the past 3 months。I cultured HCT 116 cells and counted them with the help of CASY. Then I helped Sarah with her other cell culturing and made some sections in other labs. In the second month, I practiced Western blot on PGK1 and Beta-actin. For the first time the line 3 had no trace of PGK1,
contradictory to my former results。So I figured that the concentration of proteins might be too low。In the experiment I used just 100ug（浓度，还是净含量）. I was not sure whether i had made a mistake by detecting the content of protein or else. Maybe I ought to titrate PGK1 twice and add more protein for next action。. It was not very good for Beta-actin, I couldn't find why。Sarah said this might due to less protein ,But I tested it with the R-T PCR and the result was ok.

It was not every good for Beta-actin, I did not know why, Sarah said it maybe owe to less protein, I did the R-T PCR as well and the result was ok.
during the last month, I still carried on the cell cultivation ， western blot and R-T PCR. We did western blot ，me on the beta-actin meanwhile my partner on PGK1, we used the same quantity of proteins which are different and from differing cell lines。The result on Beta-actin was unsatisfactory. By analysis I concluded we might need more protein to detect Beta-actin in western blot 。And also I sampled a lot of sections of tumors as well as nomal tissues。 I repeated R-T PCR ，although the last sample was not very satisfactory as a result of not having enough solutions supplies。
In the last month, I continued to culture cells and do western blot and R-T PCR. I did western blot on beta-actin and another person did it on PGK1, we used the same quality of protein but different protein of cells, however the result on Beta-actin was bad. By which I think we need more protein to do western blot on Beta-actin. I made a lot of sections of tumors or nomal tissues. I did R-T PCR again although the last sample is not very good for we didn't have enough solutions as we needed in our lab.
Whatelse，I have been continuing my english studies in my leisure time。I planned to take
immnohistochemical experiments after this vacation。

blue ,改成sad, blue 有忧郁的意思，但这里不合适。
blooding，应该是bleeding,bleed，动词，流血。
they couldn't embracing, 应该是embrace. 情态动词后面跟原形。
I can't stand you blooding. I only want to give you happiness." 要简单点，改成：I want to make you happy, not bleeding. 你想表达不能忍受，stand 听起来有点变味了，好像嫌弃一样。
when you was born , was 改为were.

indulgence 是纵容。 I ask the indulgence of you.，你的意思应该是，希望你原谅我？是吧，或者这样讲：“forgive me(I am so sorry或者这样简单 一点就可以了), my child, but you were born without leg.”
是个很好的故事，但是有些表达好像不太合英语的习惯，其实尽量简单就好了，听众也省力。不过没有关系啦，演讲的话，当时的发音和情感，比一些不太明显的语法错误重要得多。反正这个故事我看得明白，以上指出几点，希望帮到你，祝你成功！
